Description: Green DNA Gel Stain is a new generation of fluorescent nucleic acid gel stain designed to replace the highly toxic ethidium bromide (EtBr). The Ames test confirmed that Green Gel Stain are non-mutagenic at concentrations well above their working concentrations used for gel staining. Green Nucleic Acid Gel Stain are highly sensitive than EtBr either as precast gel stains or post gel stains. Green DNA Gel Stain has two fluorescence excitation maxima when bound to nucleic acid, at approximately 290 nm and 490 nm. The DNA bands are visible under UV light box (weak) or blue LED transilluminator (strong), which is recommended to use. Green DNA Gel Stain is a concentrated solution (10,000X in DMSO) that can be diluted 10,000 times for use in precast gel staining or ~3,300 times for use in post gel staining. The staining doesn’t affect downstream applications such as gel extraction and cloning. ProtocolPrepare a 50 ml agarose or polyacrylamide solution. Heat agarose gel solution till agarose melt completely.Add 5 μl Green DNA Gel Stain to the gel solution.Mix gently; the solution should have no air bubbles.For agarose gel, let the solution cool down to 60 - 70oC and cast the gel. For polyacrylamide gel, add APS and TEMED and cast the gel according to regular polyacrylamide gel casting protocol.Run gel electrophoresis.View the results under blue LED light.
